whats up everybody you got the car man,today we are going to be talking about a,coil in a 2000 rav4 with the,four-cylinder engine this is also,applicable in the Solera and the Camrys,with the same four-cylinder in it new,Toyota is now used the coilovers but,this is back in the day when we used,coils coil packs so whats going on with,this thing,it falls on its butt so quick we have a,check engine light on you see right,there and then after the check engine,light actually I think Ive seen my dog,stick it his nose up hes so big anyway,after the check engine light comes on,were having a serious problem with,power its falling on itself its,definitely missing and it could be a,plug wire it could be a few different,things so I went ahead and ran a scan,and lets see Ive got a p300 random,misfire that doesnt tell me much so,lets dig into this a little deeper okay,intake air ok thats because this is,unplug now all right lets just forget,that for a second now we have a cylinder,to misfire and a cylinder three misfire,we also had some air yeah system to lean,and then theres also one for the air,fuel temperature control which is also,the o2 sensor so with that being said,Im dealing with a two number two and a,number three misfire at the exact same,time and the secondary coil handles the,number two and number three cylinder so,and Im either got a blown head gasket,which I dont have any signs of that,none of that came up or its a bad coil,my bet is its a bad coil the plug wires,actually looked okay and so Im gonna,switch the switch the coil out and,thats what this video is about its,gonna be very short and sweet this is a,very simple job anyone can do it you,should not pay anyone to do this is very,simple you technically only need two,tools,three tools,[Applause],alright guys were under the hood and,youll quickly notice that the air,filter cover box is open and real simple,it just has a couple little clips on it,just flip them to the side thats it and,then here wait a minute is on the phone,whats going on here Im really bad on,flashlights thats why I end up having,to use my phone sometimes so here at the,end of this youre gonna see a little,Phillips head and basically loosen that,so you can twist this just like Im,doing the hose right here coming off the,valve cover goes right here to the side,no big deal and there is one on the,bottom you see right there literally,guys 30 seconds thats out now where is,it located,theres primary this is the one in the,four sitting right there see the spark,plugs in it and then right behind it,where the empty slot is is where the,second one goes and you see the little,white clip and theres actually a black,when the white clip goes to the front,and then theres the black one that goes,to the back now Im going to show you,how the front one is on because the back,one is exactly the same except its in,Reverse and you can actually physically,see the front one alright,okay so our were looking at the coil,here and if you look,trying to get this light there you go,that one bolt thats hanging in the back,then if you look directly on the bottom,of that thats the things I do for you,Lee you really how hard this is,[Applause],[Music],okay you can barely make out that second,one but if you were looking at this you,would see it a lot easier now all it is,is to ten millimeters they swing out and,bam its over now whats significant the,only thing youre gonna have a,difficulty is if youre not the one,thats used to changing your plugs its,how these things go but its simple,enough if you take a look its got a,little clip right here now you can just,put a little flat-head screwdriver right,under the edge of that push it out and,slip it off you dont want to put any,pressure on these wires because they,cost money to replace and you dont want,to do that because theres no reason in,it and then youll see that the cap,covers just slide freely so just pop pop,that off wiggle this off and game over,and this is the old one here assuming,this factory but I think with coil,scissors never really I mean you could,put a voltmeter on it and you know you,could test it out but thats the,original Im thinking because it has the,toyota branding on it,anyway thats it – two little ten,millimeter bolts slides in slides out,its relatively not that difficult to,get – I have really big hands and I was,able to get it so you shouldnt have a,problem,alright guys thats my quick tutorial on,how to change out one of these little,cold packs remember if you have a 1 in 4,or a 2 & 3 misfire consecutive you,probably dealing with a coil or its at,least a great place to start,excuse me meanwhile nothing but love,from the car man and the rv8 and support,your military and Ill be having another,video out to entertain you very soon,thank you so much for your support see,[Music]

this,afternoon,to me its morning i am in boston i,im sharing my screen alex can you see,my screen,yes yes all right so let me go full,screen,so before i start i wanted to thank the,organizers for the opportunity to talk,to you,today im very excited to be here thank,you for being here thank you for the,kind words alex,uh welcome to my studio actually this is,where i usually record,a podcast episodes this is actually my,closet um some podcast episodes i also,recorded in my car,as you will see uh in the next episode,that will be released soon with dr uh,brian roth so um today i am going to,talk to you about dr gpcr but uh here is,the preview,an overview of my presentation i have,three main sections,uh section number one i am going to talk,to you a little bit about geography and,history,i will run you through some of the,highlights or some of the movements,uh that determined my career as a,scientist so,i call this pre-doctoral dpcr and then i,will walk you through dr gpcr the doctor,dpc ecosystem,and the programs that weve established,to help,uh bring under the same umbrella the,whole community working on gpcrs,with the goal to better understand gpcr,biology and hopefully move forward,uh drugging gpcrs in the clinic,so a little bit of history and geography,i was born,in a city called iradia which is at the,border between hungary and romania,on the romanian side then my family and,i moved to algiers where we spent over a,decade,then moved back to romania before moving,to montreal,canada and thats where i started,my university studies at the department,of biochemistry at the university of,montreal in 2000,and that was the moment where i got,first exposed to,uh gbcrs and ill go more into details,with with these stops but my fascination,with gpcrs working three months as a,summer intern,in the lab during my undergrad led me to,a masters and a phd,also in biochemistry at the university,of montreal,in the same lab where i was able to,contribute to eight peer-reviewed,published,papers i was able to contribute also to,r d funding up to one to 1.6 million,canadian dollars and i had the,opportunity to collaborate but also to,mentor students and post docs in the lab,and not having had enough of working in,the lab and working on gpcrs i decided,to go for a post-doc,training at the rockefeller university,in new york city,and i worked in tom sachmars lab where,i was able to publish,four peer-reviewed papers i worked in,an industry collaboration so our lab had,established a collaboration with the,company,and we were able to uh obtain 400 000,in r d funding and i had the opportunity,to use my skills that i brought on from,montreal to,establish november technologies in the,lab but also mentor,students and post docs then,from new york city i moved to maryland,to bethesda,where i worked as a research fellow at,the national,institutes of health at the national,institutes of allergy and,immunology allergy,allergies and infectious diseases in the,lab of b-cell molecular immunology,its a ham its its a mouthful where,i continued working on gpcrs mainly on,on one type of gpcrs which ill get back,to later,but i also learned new techniques and i,was very,intrigued by the crispr knockout cell,line technology which i was able to,generate two crispr,and knockout cell lines and then what,you would call,going to the dark side i started working,at a biotech company,in boston where its still located so,the company,um is called generate biomedicines,formerly called,the l56 for ventral lab 56,where i used my experience working on,gpcrs and my scientific knowledge,to further um work on a family of gpcrs,and the company focused on using,ai and machine learning to explore the,protein space and im going to get back,to uh to this a little bit later,and then covet hit and it was a blessing,in disguise,and um in march 2000 2020 this year i,decided that i wanted to do something,different,and i founded dr gpcr and within dr gpc,are various programs,to help scientists working on in the,field connect,so one of these programs is my podcast,that alex,also had talked about so so far we,i recorded 30 podcast episodes with,various um various guests we released 13,episodes,so far we have over 2 000 listeners,for this app for for the podcast uh we,also have a newsletter a monthly,newsletter that goes out to 600,recipients,many many twitter followers linkedin and,facebook followers and i want to thank,them for that,and then i think the highlight of dr,gbcr for this year,was organizing the dr gpcr summit with,30 phenomenal,various talks and 400 registrants for,the summit,so now im going to take you through,snippets of these different stops that i,had through my career,and im going to highlight some some of,the lessons that i learned,during that time so my first contact,with gpcrs happened at the university of,montreal the department of biochemistry,and it was a class of uh of about,signalling pathways,and dr michel bouvier was teaching the,class,i dont specifically remember what he,was talking about i was just fascinated,by,how many signaling pathways could be um,activated when you when a receptor was,binding its its ligand,so during this training,during this this degree undergraduate,degree there was an opportunity to spend,the summer,in a research lab and out of,out of uh disappointing news that,michelle didnt couldnt have me,work in his lab he had offered the,position to one of my,best colleagues and good friends he,introduced me,to someone named nicolas ivaker who uh,became actually my mentor,at the san jose hospital research center,and nicolas at the time had opened his,lab so when i,entered the lab for an interview there,was no equipment there were only,bills everywhere on every desk and every,bench so i was the first,student in the lab and nicolas was,working on chemokines and chemokine,receptors,and as soon as i heard about the,chemokines and chemokine receptor system,i thought it would it was a very,interesting and very cool system to work,about,uh work on
